Description
DiR is a lipophilic, near-infrared fluorescent cyanine dye. The dye is useful for labeling cytoplasmic membranes. The two long 18-carbon chains insert into the cell membrane, resulting in specific and stable cell staining with no or minimal dye transfer between cells.
DiR (near infrared fluorescence) and other cell membrane fluorescent dyes such as DiI (orange fluorescence), DiO (green fluorescence), DiD (red fluorescence) are used together to provide an effective tool for multicolor imaging and flow cytometry.
Cells can be fixed with formaldehyde either before or after staining, but not recommended for permeabilization.
Notes
1. When DiR stains fixed cell or tissue samples, the samples should be fixed with 4% paraformaldehyde in PBS. The use of other improper fixing solutions will result in a high fluorescence background.
2. There are quenching problems with fluorescent dyes. Please avoid light to slow down the fluorescence quenching.
