Description
Annexin V (membrane-linked protein-V) is a Ca2+-dependent
phospholipid-binding protein with a molecular weight of 35-36 KD that binds
selectively to phosphatidylserine (PS). PS is mainly distributed on the inner
side of the cell membrane, i.e., the side adjacent to the cytoplasm. In the
early stages of apoptosis, different cell type exocytose phosphatidylserine to
the cell surface and are exposed to the extracellular environment. At this
point, the use of fluorescent protein PE-tagged Annexin V, Annexin V-PE, in
combination with the outside-out PS allows flow cytometry to directly detect
the outside-out PS, an important feature of apoptosis. Normal cells are not
stained by Annexin V-PE, cells undergoing apoptosis or necrosis are stained by
Annexin V-PE. Annexin V-PE can be used in combination with partially
non-permeable nuclear dyes (7-AAD/PI) to differentiate cells in
different periods of apoptosis.
RedNucleus II provided in this kit is a Far-infrared fluorescence dye,
belonging to anthraquinone compounds, which cannot penetrate the intact cell
membrane of living cells and early apoptotic cells.It is impermeable, but can
rapidly stain nucleus/dsDNA in dead and permeable cells. Rednucleus II is an
ideal substitute of propidium iodide (PI) and 7-AAD. It can be used in
combination with Annexin V-PE without compensation adjustment and has better spectral characteristics: it is not excited by ultraviolet rays, nor
does it interact with PE/PE. The homologues have overlapping emission,
so it can be combined with FITC, PE and violet fluorescent dyes for multi-color
analysisIt. When combined with Annexin V-PE, RedNucleus II is excluded from
living cells and early apoptotic cells, while late apoptotic cells and dead cells
are stained with Annexin V-PE and RedNucleus II at the same time.
Annexin V-PE and RedNucleus II Apoptosis Kit can be detected by flow
cytometry or other fluorescence detection equipment.
